Coffee is one of the most valuable agricultural commodities and ranks second on international trade exchanges. The genus Coffea belongs to the Rubiaceae family which includes other important plants. The genus contains about 100 species but commercial production is based only on two species, Coffea arabica and Coffea canephora that represent about 70 % and 30 % of the total coffee market, respectively. The Brazilian Coffee Genome Project was designed with the objective of making modern genomics resources available to the coffee scientific community, working on different aspects of the coffee production chain. Caracterização molecular de animais da raça Nelore utilizando microssatélites e genes candidatos R. Bras. Zootec.

No presente trabalho, 180 fêmeas da raça Nelore, provenientes de oito rebanhos, foram analisadas quanto aos marcadores microssatélites TEXAN15, BM1224 e CSFM50 e aos polimorfismos de comprimento de fragmentos de restrição (RFLP) nos locos k-caseína, beta-lactoglobulina e hormônio de crescimento (GH). Com exceção de GH, todos os marcadores foram polimórficos na amostra estudada. Os valores de heterozigosidade, diversidade gênica, conteúdo de informação polimórfica (PIC) e probabilidade de exclusão de paternidade (PE) foram estimados. Os maiores valores de PIC (0,685) e PE (0,521) foram obtidos para o marcador BM1224.

Cross-species microsatellite amplification in South American Caimans (Caiman spp and Paleosuchus palpebrosus) Genet. Mol. Biol.

Microsatellite DNA markers have been used to assess genetic diversity and to study ecological behavioral characteristics in animals. Although these markers are powerful tools, their development is labor intensive and costly. Thus, before new markers are developed it is important to prospect the use of markers from related species. In the present study we investigated the possibility of using microsatellite markers developed for Alligator mississipiensis and Caiman latirostris in South American crocodilians. Our results demonstrate the use of microsatellite markers for Paleosuchus palpebrosus, Caiman crocodilus and Caiman yacare.

Este trabalho foi realizado com o objetivo de avaliar o efeito da adição de ractopamina na ração sobre a qualidade da carne de suínos dos genótipos halotano homozigoto dominante (HalNN) e heterozigoto (HalNn). Durante o experimento (21 dias), 24 suínos machos castrados e 12 fêmeas (metade de cada genótipo) com peso médio inicial de 72,6 kg de PV foram avaliados segundo delineamento experimental em blocos casualizados, em arranjo fatorial 2 x 2 x 2 (dois genótipos halotano; duas rações: controle e com adição de 10 ppm de ractopamina; e dois sexos: machos castrados e fêmeas). Aberdeen Angus beef cattle from the Brazilian herd were studied genetically using restriction fragment length polymorphism (RFLP) of the kappa-casein - HinfI (CSN3 - HinfI), beta-lactoglobulin - HaeIII (LGB - HaeIII) and growth hormone AluI (GH- AluI) genes, as well as four microsatellites (TEXAN15, CSFM50, BM1224 and BM7160). A genetic map provides insight into genome organization and chromosomal location of markers and genes, and is important for quantitative trait loci (QTL) mapping. The objective of this study was to use a Brazilian resource population to construct a linkage map for chicken chromosome 1. Eighty microsatellite markers were tested and 26 informative markers were typed in an experimental F2 population developed by two generations of crossbreeding between a broiler sire line and a layer line. A linkage map is essential not only for quantitative trait loci (QTL) mapping, but also for the organization and location of genes along the chromosomes. The present study is part of a project whose major objective is, besides from construction the linkage maps, the whole genome scan for mapping QTL for performance traits in the Brazilian experimental chicken population. Chicken embryos were incubated with BrdU, embedded in plastic resin, sectioned and screened immunohistochemically to identify proliferating cells in the neural tube and somites. Fixation in 4% paraformaldehyde for 1 h was essential for detecting specific colorimetric signals of BrdU incorporation into cells during the S phase of the cell cycle. Transverse sections of the neural tube showed that the nuclei of proliferating cells (BrdU positive) had a uniform and centralized distribution, whereas unstained nuclei were found only along the extremities of the neural tube. This study aimed at mapping QTL (quantitative trait loci) using linear combinations of characteristics of economical interest in Gallus gallus. A total of 350 F2 chickens from an initial crossing among males from a broiler line (TT) with females from a layer line (CC) were used. Molecular cloning of αRYR hotspot region 1 from broiler chicken BABT

Samples of Pectoralis major m. were collected, and an RT-PCR analysis of the a-Ryanodine receptor (a RYR) from chicken mRNA hotspot region spanning aminoacid residues 386 to 540, numbered according to the turkey sequence, revealed two classes of transcripts. The sequences of the first class were similar to turkey and human with 97% and 74% of identity, respectively, and included all transcripts with substitutions in the nucleotide sequence. The second class was characterized by the deletion of nucleotides, leading to a premature stop codon and coding for a truncated and nonfunctional protein. Several studies have investigated the relationship between heterozygosity, genetic distance and production traits. The objective of the present study was to evaluate the influence of the degree of heterozygosity and genetic distance on growth, carcass and reproductive related features in F1 bovine crosses. We tested 10 polymorphic markers in 330 purebred cattle (Nelore, Canchim, Aberdeen Angus and Simental) and 256 crossbred cattle belonging to four crossbred groups. Abstract: The objective of this work was to sequence the MyoD1, MyoG, MyF5, MyF6, and MSTN genes and to identify polymorphisms in Santa Inês sheep (Ovis aries). A total of 192 lambs with 240 days of age were evaluated, and these genes were sequenced to be compared with the reference sequence in the Ovis aries genome. Genotype and allele frequencies were estimated, and the Hardy-Weinberg equilibrium was tested. Fragments containing 2,493 bp (MyoD1), 1,836 bp (MyoG), 2,813 bp (MyF5), 1,126 bp (MyF6), and 2,380 bp (MSTN) were obtained, and, in these sequences, 160 variants were identified. Quantitative trait loci (QTL) mapping in livestock allows the identification of genes that determine the genetic variation affecting traits of economic interest. We analyzed the birth weight and weight at 60 days QTL segregating on bovine chromosome BTA14 in a F2 resource population using genotypes produced from seven microsatellite markers. Phenotypes were derived from 346 F2 progeny produced from crossing Bos indicus Gyr x Holstein Bos taurus F1 parents. Segregation between a genetic marker and a locus influencing a quantitative trait in a well delineated population is the basis for success in mapping quantitative trait loci (QTL). To detect bovine chromosome 5 (BTA5) birth weight QTL we genotyped 294 F2 Gyr (Bos indicus) x Holstein (Bos taurus) crossbreed cattle for five microsatellite markers. A linkage map was constructed for the markers and an interval analysis for the presence of QTL was performed.
